摘要:
【目的】探索不同碳源对耐热型解脂亚罗酵母生长及产赤藓糖醇的影响,通过优化碳源组合提升耐热型解脂亚罗酵母的赤藓糖醇产量。【方法】分别以30、34、38 ℃条件下生长的解脂亚罗酵母CA20、解脂亚罗酵母HT34和解脂亚罗酵母HT38为研究对象,利用含有不同碳源的培养基进行培养。培养过程中每隔24 h取样,采用酶标仪检测菌液在600 nm处的光密度值(optical density, OD600)。培养液中赤藓糖醇和葡萄糖质量浓度采用高效液相色谱(HPLC)检测。【结果】菌株CA20能利用葡萄糖、果糖、半乳糖、阿拉伯糖、木糖、麦芽糖、蔗糖、纤维二糖、甘露醇、山梨醇、甘油、可溶性淀粉和大豆油脂等碳源,其中葡萄糖、果糖、甘露醇、山梨醇、甘油和大豆油脂为最优碳源; 菌株HT34对除葡萄糖和果糖以外碳源的利用能力下降,而菌株HT38几乎不能利用除葡萄糖和大豆油脂以外的碳源; 培养基中同时添加葡萄糖和另一种碳源对菌株CA20生长的影响较小,而同时添加葡萄糖和大豆油脂可明显促进菌株HT34和HT38的生长。此外,大豆油脂可显著提升菌株HT34和HT38的赤藓糖醇产量和得率,增加量最高可达50%左右。【结论】本研究结果为今后利用高温发酵生产赤藓糖醇提供了理论依据。
